●WWE Domain (RNF146) Poly-ADP-ribose Affinity Resins

The Tulip Biolabs, Inc. RNF146 WWE Domain Affinity Resins are designed for the isolation of poly-ADP-ribosylated (PARylated) proteins from cultured cells and tissue lysates.

RNF146 (Iduna) is a RING-domain E3 ubiquitin ligase that positively regulates Wnt signalling.  RNF146 directly interacts with poly(ADP-ribose) through its WWE domain.  The WWE domain is a conserved globular domain found in multiple PARPs and E3 ligases.  Through the use of WWE domain affinity resins, poly-ADP-ribosylated (PARylated) proteins can be isolated from cell or tissue lysates.  The resin bound proteins can be eluted from the affinity resin, and analyzed by immunoblotting, mass spec, and other methods.  Tulip Biolabs produces two types of WWE domain beads, see below:

WWE Negative Control Resin (agarose beads)

Description: Identical to the #2334 affinity resin except for a single amino acid substitution R163A, which abolishes PAR binding.  The negative control resin is useful as a control for non-specific binding.

Supplied as: 0.5mg protein/0.5mL buffer as a suspension containing ~50µL resin.

WWE Domain Magnetic Affinity Resin (covalent)

Description: Cat. #2438 WWE Domain Magnetic Resin is highly purified WWE RNF146 domain fusion protein expressed in E. coli, and covalently bound to superparamagnetic silica-based beads (resin). Typical conditions for the elution of the affinity bound proteins to the resin, for example SDS-PAGE sample buffer, will not dissociate the WWE domain affinity protein from the resin.  The magnetic resin beads are small, with an average 1µm diameter, which remain suspended in solution for a few minutes after mixing, allowing for convenient and accurate pipetting.  Tulip Biolabs Cat. #2334 (non-magnetic) WWE domain agarose beads, above, are composed of the same affinity protein except non-covalently bound to glutathione beads.  The resin is useful for affinity purification (pulldown) of PARylated proteins.

Negative Control Magnetic Resin (covalent)

Description: Cat. #2427 Negative Control Magnetic Resin are the same magnetic beads used to produce the #2414, 2426, and 2438 mag affinity resins, but instead of attaching the corresponding affinity protein to the resin, the resin is modified to an alpha amino carboxylic acid. Use equivalent volume of the 2427 neg control mag resin as experimentally used for the 2414, 2426, or 2438 mag affinity resins as a control for non-specific binding to the resins.

Supplied as: Each vial contains 0.5mL Negative Control magnetic resin beads in approximately equivalent quantity as the 1mg/mL mag affinity resins 2414, 2426, and 2438.
